One of the most fundamental barriers of using light in biomedicine is the inherent trade-off between spatial resolution and penetration depth due to the strong scattering of light caused by the inhomogeneities of refractive index in biological tissue. Breaking this limitation has long been the dream of the community, but recent progress in wavefront engineering, including optical phase conjugation and wavefront shaping, has brought new hopes to it. Especially, when noninvasive guide stars, such as ultrasonic mediation, are employed, it has been demonstrated that diffused light can be focused tightly to wherever the guide stars with an acoustic- or even optical diffraction-limited resolution in deep biological tissue. In this talk, we will describe such examples of using two types of ultrasonic mediation, i.e., ultrasound-modulation of light and photoacoustic sensing, as the guide star for optical phase conjugation and wavefront shaping, respectively. Despite of the limitations in either focusing speed or efficiency, with further improvements in the field, reshaping light controllably at depth in living tissue is on the horizon, which may potentially benefit a wide range of optical applications, including imaging, sensing, therapy, controlling, and stimulation, that prefer high-resolution and high-contrast optical delivery in deep tissue. The potential impact is limited only by our imagination.

Dr. Puxiang Lai received his Bachelor of Engineering in Biomedical Engineering from Tsinghua University, China in 2002, Master of Science in Acoustics from the Chinese Academy of Sciences, China in 2005, and PhD in Mechanical Engineering from the Boston University, USA in 2011. After that, Dr. Lai joined Dr Lihong Wang’s Optical Imaging Lab in the Washington University in St. Louis as a Postdoctoral Research Associate. In September 2015, he joined the Department of Biomedical Engineering at the Hong Kong Polytechnic University as tenure-tracked Assistant Professor.
Dr Lai’s research focuses on the development of novel biomedical imaging, therapy, and manipulation modalities by using light and sound, including (but are not limited to): 1) noninvasive imaging techniques, such as ultrasonic imaging, acousto-optic imaging, as well as photoacoustic/optoacoustic imaging; 2) focused ultrasound therapy and its real-time monitoring; 3) optical speckles; and 4) deep-penetrating optical focusing, imaging, sensing, stimulation and control. These studies have served to substantially advance the state of the art in biomedicine, and made Dr Lai being recognized within and outside his field. For example, his research has thus far fuelled more than 25 top journal publications (including two first-authored in Nature Photonics and Nature Communications, respectively), more than 55 invited or contributing talks at important international conferences and winning several best paper/poster awards. Dr Lai has also been invited to give seminars in many famous universities and research institutions, and served as reviewer or editor for more than 25 premium journals. Since joining BME at PolyU, Dr. Lai aims to continue his research on deep-tissue probing and treatment by using light and sound, which has been supported by the Hong Kong Polytechnic University, the Hong Kong Research Grants Council (RGC), and the National Natural Science Foundation of China (NSFC). Recently, he was awarded the 2016 Hong Kong RGC Early Career Award.
